Job Description SENIOR RESEARCHER

Abbott Laboratories in Lake County, Illinois seeks a qualified senior researcher in the area of drug metabolism with responsibilities for design and conduct of in vitro and in vivo drug transport studies. PhD in Pharmaceutical Sciences or related field such as Pharmacology and Pharmacy required with 2 years of post-doctoral experience in a drug transporter related field in an academic and/or industrial environment, including: (i) Characterization/prediction of drug elimination pathways in the liver, kidney and brain requiring conduct of suitable in vitro transporter experiments such as uptake assays, transcellular transport assays, vesicle transport assays and ATPase assays using transfected cell lines and membrane vesicles, and the use of radiochemicals and analytical techniques including LC-MS/MS in particular; (ii) Assessment/prediction of potential drug-drug interaction mediated by transporters by evaluating drug transporter kinetics, and; (iii) Implementation of state-of-art technologies in the field of drug transporters, requiring hands-on experience in molecular biology and biochemical techniques including cell culture, recombinant DNA technology, gene induction assays, Western blot and immunohistochemistry in a research-oriented environment. Responsible for the design & conduct of detailed in vitro & in vivo drug transport studies to evaluate the transport & kinetics of discovery & development drug candidates in order to elucidate mechanisms & route of drug elimination from the kidney, liver & brain: (I) Elucidation of Drug Efflux Pathways Using In Vitro Systems: Characterization of drug elimination pathways in the liver & kidney & investigation of the contribution of drug transporters to the elimination of test compounds from the brain across the blood-brain barrier incl.: (i) development & conduct of suitable in vitro transporter experiments to measure & predict pathways of drug elimination from the liver, kidney, & brain; (ii) uptake & transcellular transport assays using transporter-overexpressing (transfected) mammalian cell lines (e.g. human embryonic kidney (HEK) & Madin-Darby canine kidney (MDCK) cells), & vesicle transport & ATPase assays using membrane vesicles expressing efflux transporters, which require handling of radiochemicals & analytical techniques such as liquid chromatography-tandem mass spectrometry (LC-MS/MS), &; (iii) calculation of relevant drug concentrations & design of approp. incubation conditions; (II) Elucidation of Renal Drug Uptake & Prediction of Drug-Drug Interactions Mediated by Transporters: Prediction/determination of the involvement of drug transporters in the active renal (i.e. kidney) secretion &/or reabsorption of preclinical drug candidates & in drug-drug interactions, requiring: (i) the conduct of studies w/HEK cell lines expressing organic anion or cation transporters to measure the uptake of test compounds into those cell lines & investigate the inhibitory effect of test compound on the uptake of probe substrates w/quantitative analysis using radiochemicals &/or LC-MS/MS; (ii) the assessment & prediction of potential drug-drug interaction mediated by transporters using relevant in vitro/in vivo information incl. clinical data, & evaluation of the drug transporter kinetics to support clinical development of drug candidates, &; (iii) the calculation of relevant drug concentrations & design of approp. incubation conditions; (III) Application & Development of Transporter Methods: Improve current techniques in drug transport & influence the internal & external scientific environment by implementing new state-of-art technologies in the field of drug transporters & by providing expert scientific & technical advice based on current literature & opinion leaders, requiring: (i) a broad wealth of exp. & knowledge in the design & conduct of drug transport studies, &; (ii) hands-on exp. in molecular biology & biochemical techniques incl. cell culture, recombinant DNA technology, gene induction assays (e.g. Luciferase assay), Western blot, & immunohistochemistry in a research-oriented environment, &; (IV) Scientific Report Writing & Scientific Presentation: Write internal technical scientific reports & memos/reports for regulatory submissions, communicate scientific findings to multi-disciplinary project teams, & interpret, evaluate, review & summarize drug metabolism & pharmacokinetics reports from internal & external sources, requiring: (i) an understanding of the drug metabolism & pharmacokinetics studies executed; (ii) excellent scientific writing skills in the area of drug metabolism & pharmacokinetics as reflected by a strong record of scientific publications of the candidate & abilities to communicate research findings to multi-disciplinary project teams &/or regulatory agencies via report writing & oral communication, &; (iii) capability to engage in discussion & decisions on materials outside the own area of expertise.
