To answer your question, of course they do. It's part of their more extensive background check. But if you "accidentally" put down a different GPA on your application than what it really is, unless you lied so badly that you really tried to pull one over on them, they won't make an issue of it. If the hiring manager wants you, he/she will find a way.
But if your GPA slip is part of a bigger string of lies, you can guarantee it will be an issue.
